MP is 68/12 over 51 hands.
BU is 48/3 over 32 hands.
BB is 70/40 over 32 hands.

First of all, knowing there's two loose players behind me, is it fine to isolate on the fish here in position?

Second, what is the best plan on the river. I was going for a bet/fold against BB and MP, but a bet/call against BU because of his stack size.

I would raise TK to get value from button and the fish. Our range is ahead of their calling range so it's fine. I'll fold it usually though.

Against fishes with top pair good kicker I think we can keep betting for value all streets. However when bu jams 4 way I'm actually inclined to fold. If he jammed heads up his range would be weaker but 4 way I think he has more strong made hands than draws and we can fold TK
