Do I first have to upload Moneybookers with my own money (bank transfer) before I can send money to other sites?
Or can I ask a friend to send me some money from his MB account, so I can send that to a poker site?

Yes, friends can send you, but I advise you to upload some small sum from your bank too.
This way you will have your account verified and you won't have any problems in future.
MB "froze" my outgoing transaction until I deposited my own funds. Since then everything is working like a charm.
Also your "transfer" limit will be higher.
