depends on what you feel comfortable with... If your feel confident sat with 250BBs at the table then it will be an advantage should you manage to get all in with a monster, on the flip side, Bad beats will be alot harder to take, should it be against another 250BB deep player.

i have no problem sitting with 250BB and not many are buy in with that from what i see
Your bankroll = the amount of money you set aside to play poker:
If you have $300 on site A and $50 on site B then your bankroll is $350. Feel free to jump right into NL10 or whatever you are comfortable playing.
With 5 $ buy in you have only 10 BI with 2 $ buy in you have 25 . IMO play with 2 $ buy in until you reach 100 $ Bankroll
